Alzheimer's Vaccine Draws Closer To Human Trials

    https://www.utsouthwestern.edu/newsr...lzheimers.html

DALLAS – Nov. 20, 2018 – A DNA vaccine tested in mice reduces accumulation of both types of toxic proteins associated with Alzheimer’s disease, according to research that scientists say may pave the way to a clinical trial.

Above: A DNA vaccine given to a mouse modeled to have Alzheimer’s disease reduced beta-amyloid plaques (red) and tau tangles (green) in the hippocampus region of the brain. Below: An untreated mouse modeled to have Alzheimer’s disease accumulated notably more beta-amyloid plaques (red) and tau tangles (green) in the hippocampus region of the brain.
A new study by UT Southwestern’s Peter O’Donnell Jr. Brain Institute shows that a vaccine delivered to the skin prompts an immune response that reduces buildup of harmful tau and beta-amyloid – without triggering severe brain swelling that earlier antibody treatments caused in some patients.

“This study is the culmination of a decade of research that has repeatedly demonstrated that this vaccine can effectively and safely target in animal models what we think may cause Alzheimer’s disease,” said Dr. Roger Rosenberg, founding Director of the Alzheimer’s Disease Center at UT Southwestern. “I believe we’re getting close to testing this therapy in people.”

The research published in Alzheimer’s Research and Therapy demonstrates how a vaccine containing DNA coding for a segment of beta-amyloid also reduces tau in mice modeled to have Alzheimer’s disease. In addition, the vaccine elicits a different immune response that may be safe for humans. Two previous studies from Dr. Rosenberg’s lab showed similar immune responses in rabbits and monkeys.

The vaccine is on a shortlist of promising antibody treatments aimed at protecting against both types of proteins that kill brain cells as they spread in deadly plaques and tangles on the brains of Alzheimer’s disease patients.

Although earlier research established that antibodies significantly reduce amyloid buildup in the brain, Dr. Rosenberg’s team needed to find a safe way to introduce them into the body. A vaccine developed elsewhere showed promise in the early 2000s, but when tested in humans, it caused brain swelling in some patients.

Alzheimer’s funding tops $1.9 billion annually; more needed

PHOENIX — Federal research funds that could prevent or lead to the cure for Alzheimer’s disease have reached $1.9 billion annually but still lag far behind research money for cancer and HIV/AIDS, experts said.

Funds from the National Institutes of Health, the federal research agency that funds grant proposals, tripled from 2015 to 2018. By comparison, cancer research funding reached $6.6 billion.

Alzheimer’s disease is the least funded chronic disease in the U.S., according to James Fitzpatrick, director of programs and advocacy at Desert Southwest Chapter of the Alzheimer’s Association.
“Cancers and HIV and other diseases that are in the top five in our country are still receiving way more than what we are, and it’s not enough,” Fitzpatrick said.

Fitzpatrick said that until the first baby boomer turned 65 in 2011, Alzheimer’s disease hadn’t reached the diagnosis levels now reaching that generation. Awareness started to ratchet up after former President Ronald Reagan was diagnosed with Alzheimer’s in 1994.

“It didn’t really hit home to society until Reagan came out with the disease, and since then there has been an incline for research funding, but not as huge an incline as it has been the last four years,” he said.

For Alzheimer’s to receive more federal funding, Fitzpatrick said, it has to come out of the research budget for other diseases, which presents a dilemma.

    A path toward a cure

Edward Ofori, an Arizona State University professor, said more funds would increase the chances of finding a cure for a disease that as yet has no survivors.

I don't mind losing my mind, but I don't want to go into a nursing home simply because I can't be trusted with a stove.

Hopefully in 50 years DonnaY's grandchildren will post articles claiming that it can be cured with diet and/or it disappeared naturally and the vaccines are poison.

I am sure that vaccine is loaded with the commonly used vaccine adjuvant--aluminum. No thanks.

"Many researchers are beginning to accept that aluminum has some sort of role to play in neurodegenerative diseases such as Alzheimer's. Whether it does in others is still an open question, but Alzheimer's is really coming into focus and it's fairly clear that the body burden of aluminum from all the sources to which humans are exposed may be contributing to Alzheimer's disease." ~ Christopher Shaw - neuroscientist.

34 studies were selected, among which 68% established a relation between Al and AD, 23.5% were inconclusive and 8.5% did not establish a relation between Al and AD.

    2011 https://www.ncbi.nlm.nih.gov/pubmed/21157018
The hypothesis that Al significantly contributes to AD is built upon very solid experimental evidence and should not be dismissed. Immediate steps should be taken to lessen human exposure to Al, which may be the single most aggravating and avoidable factor related to AD.

    2011 https://www.ncbi.nlm.nih.gov/pmc/articles/PMC3056430/
There is growing evidence for a link between Al and AD, and between other metals and AD.

    2013 https://www.ncbi.nlm.nih.gov/pubmed/23380995
Many humans routinely ingest aluminum salts as additives contained in processed foods and alum-treated drinking water. The physical properties of aluminum and ferric iron ions are similar, allowing aluminum to use mechanisms evolved for iron to enter vulnerable neurons involved in AD progression, accumulate in those neurons, and cause neurofibrillary damage.

    2014 https://www.ncbi.nlm.nih.gov/pmc/articles/PMC3986683/
We think it important to emphasize that the most effective clinical treatment yet devised for moderate- to late-stage AD patients was the implementation of the first generation anti-oxidant and trivalent iron/aluminum chelator desferrioxamine to attempt to remove aluminum from the brains of AD patients

    2014 https://www.ncbi.nlm.nih.gov/pmc/articles/PMC3986683/
Aluminum's contribution to AD is based upon at least seven independently derived observations:

    2014 https://www.ncbi.nlm.nih.gov/pubmed/24577474
AD is a human form of chronic aluminum neurotoxicity. The causality analysis demonstrates that chronic aluminum intake causes AD.

    2016 https://www.ncbi.nlm.nih.gov/pubmed/26592479
(with a total of 10567 individuals) Results showed that individuals chronically exposed to Al were 71% more likely to develop AD (95% confidence interval)

    2017 https://www.ncbi.nlm.nih.gov/pubmed/28159219
The unique quantitative data and the stunning images of aluminium in familial Alzheimer's disease brain tissue raise the spectre of aluminium's role in this devastating disease.

Further studies of symbiotic HM-CNS communication intrinsically suggests the potential for microbial-based therapeutic strategies that may aid in the augmentation of the HM, for the treatment of human disease, including neurological disorders

All the results suggest that AD may begin in the gut, and is closely related to the imbalance of gut microbiota.

Thus, modulation of gut microbiota (by probiotics or other dietary intervention) or direct targeting of gut microbiota enzymes (by pharmacological inhibitors or activators) may be a growing area for pharmaceutical and functional food industries, with the goal of decreasing the widespread growth of adiposity, insulin resistance, AD, and other metabolic diseases.

Our studies support the hypothesis that intestinal microbiota may help protect against AD, in part, by supporting the generation of select SCFAs, which interfere with the formation of toxic soluble Aβ aggregates

5 neurotoxins found in your favorite foods

Friday, November 23, 2018 by: Ellaine Castillo

Nowadays, people have become so busy that they don’t even have time to prepare fresh food. Many have become dependent on processed and fast foods because of their convenience. However, many of these foods contain additives that are still allowed by the U.S. Food and Drug Administration even though they are known neurotoxins.

Neurotoxins are naturally occurring or artificially produced substances that have damaging effects on the nervous system. Overall, there are more than 1,000 chemicals that have been identified as neurotoxins. Although most of these are derived from external sources like food, there are also some that are produced by the body. Exposure to these neurotoxins can lead to cognitive and behavioral problems as well as disorders like autism and attention deficit hyperactivity disorder (ADHD).

Most consumers trust that these harmful substances won’t be found in food. Unfortunately, this isn’t the case. Lots of foods available on the market contain neurotoxic additives to improve flavor and make people want them more. Some of the most common neurotoxins that you can find in processed foods include the following:

    Aspartame
This artificial sweetener is commonly found in diet sodas and other “sugar-free” foods. It is composed of aspartic acid, phenylalanine, and methanol, which are also known to inflict damage on the brain. Aspartame intake can cause various side effects, which include migraines, dizziness, anxiety, memory loss, Alzheimer’s, and fibromyalgia. Moreover, a previous study has also shown that soft drinks that contain this neurotoxin increase the risk of depression.

If you want an alternative to sugar and aspartame, then you can try using stevia (Stevia rebaudiana). This herb is excellent for those who are trying to lose weight because it has zero calories. Moreover, it also doesn’t increase blood sugar or cause tooth decay.

    Diacetyl
Diacetyl is a common additive in the butter flavoring of microwave popcorn. This substance is especially harmful since it can cross the protective blood-brain barrier. Once it reaches the brain, it causes beta-amyloid plaques to form, which puts the person at risk of Alzheimer’s. Making popcorn the old-fashioned way is a great way of avoiding diacetyl since you can flavor it with real butter.

    Monosodium Glutamate (MSG)
Free glutamate, like MSG, is especially harmful to the body since both are rapidly absorbed into the bloodstream. Although glutamate is necessary for your brain to function, too much of it can overstimulate brain cells to the point where they start dying off. Moreover, MSG intake can increase the risk of psychological disorders and neurodegenerative diseases.

    Mercury
Unlike the previous neurotoxins, mercury is not deliberately added to food. This compound finds its way to fish and other seafood through pollution in the different bodies of water. Mercury tends to build up in the brain, but it takes some time for it to cause toxicity, which could manifest as memory loss, depression, anxiety, mood swings, numbness, or tremors. Since fish are excellent sources of omega-3 which is beneficial for the brain, it’s not good if you completely avoid them. You just have to choose the ones that are low in mercury, which include Alaskan salmon, sardines, herring, and mackerel.

    Aluminum
Aluminum is found in baking powder, anti-caking agents, drinking water, antacids, and even in food packaging materials like cans and foil. Previous studies have shown that this element increases Alzheimer’s risk by up to 71 percent. Fortunately, there are aluminum-free products available on the market that you can use to reduce exposure.

By familiarizing yourself with the neurotoxins that are commonly added to your food, you can become a smarter and healthier consumer.

There is evidence that aluminium and fluoride cause dementia, Alzheimer's disease...
Maybe you disagree that aluminium should be removed from vaccines?

The following study shows that vitamin B9 (a.k.a. folate, folic acid and folacin) significantly reduces the risk of dementia.
